On September 1, 2026, Alumis Inc. announced that its Phase 2b LUMUS trial of envudeucitinib for systemic lupus erythematosus (SLE) failed to meet primary and secondary endpoints in the overall population of 408 patients. The failure was attributed to a higher-than-expected proportion of IFNGS-low patients, which drove high placebo response rates. However, robust clinical responses were observed in the prespecified IFNGS-high subgroup across BICLA, SRI-4, and CLASI-50 endpoints. Pharmacodynamic data confirmed dose-dependent interferon-pathway target engagement with no new safety signals. Alumis plans to engage regulators regarding Phase 3 development for this specific subgroup while remaining on track to submit a New Drug Application (NDA) for envudeucitinib in moderate-to-severe plaque psoriasis in the fourth quarter of 2026. Alumis Inc. is a late-stage biopharmaceutical company developing targeted therapies for immune-mediated diseases.
