On June 15, 2026, the US FDA approved Truqap (capivasertib) in combination with abiraterone and prednisone as the first targeted treatment for adult patients with PTEN-deficient metastatic androgen pathway modulation-naïve or sensitive (mAPMN/S) prostate cancer. The approval follows the Phase III CAPItello-281 trial, which demonstrated a 19% reduction in the risk of radiographic disease progression or death (HR 0.81) and improved median radiographic progression-free survival to 33.2 months compared to 25.7 months for the control group. The FDA also approved a companion diagnostic test to detect PTEN deficiency. A similar regulatory application is currently under review in the EU.
