AstraZeneca and Daiichi Sankyo announced positive high-level results from the Phase III DESTINY-Lung04 trial, demonstrating that Enhertu (trastuzumab deruxtecan) significantly improved progression-free survival compared to global standard of care as a first-line treatment for patients with HER2-mutant advanced non-small cell lung cancer. The trial enrolled 454 patients and showed no new safety concerns beyond the drug's known profile. This marks the first Phase III trial to demonstrate such a benefit in this setting, potentially allowing Enhertu to move earlier in the treatment algorithm for an aggressive cancer with historically limited targeted options. AstraZeneca is discontinuing the eVOLVE-Lung02 Phase III trial of volrustomig plus chemotherapy for metastatic non-small cell lung cancer (mNSCLC) with PD-L1 <50%. The Independent Data Monitoring Committee recommended termination, concluding the combination was unlikely to meet primary endpoints for progression-free or overall survival in patients with PD-L1 negative tumors (<1%). The trial enrolled 895 patients across 25 countries. Other Phase III volrustomig trials in cervical cancer, head and neck squamous cell carcinoma, and mesothelioma continue as planned. AstraZeneca reported positive high-level results from the global Phase III SAFFRON trial, demonstrating that Tagrisso (osimertinib) plus Orpathys (savolitinib) significantly improved progression-free and overall survival compared to platinum-based chemotherapy in patients with EGFR-mutated non-small cell lung cancer who had progressed on prior Tagrisso treatment. The combination targets MET-driven resistance, a common mechanism of disease progression in this population. These findings reinforce the potential for global regulatory approval and commercialization of the combination therapy, which is already approved in China based on the SACHI trial.
